| 背景信息 | Tripartite motif containing 8 (TRIM8) is an E3 ubiquitin-protein ligase that regulates innate immunity, inflammation, and cell cycle progression. It features a classic TRIM structure that supports diverse ubiquitination activities, including K63- and K48-linked chains. TRIM8 promotes NF-κB and AP-1 activation by mediating the K63-linked polyubiquitination of TAK1. It also acts as a tumor suppressor by stabilizing p53 through the disruption of its interaction with MDM2, thereby promoting DNA repair and cell cycle arrest. Additionally, TRIM8 modulates STAT3 signaling and supports chromosomal stability through interactions with centrosomal proteins like Plk1. Clinically, dysregulation of TRIM8 is implicated in various cancers, including colorectal, thyroid, and breast carcinomas, where it influences tumor stemness and chemotherapy sensitivity.
