| 背景信息 | Tripartite motif containing 2 (TRIM2) is a UbcH5a-dependent E3 ubiquitin ligase belonging to the TRIM-NHL family, primarily expressed in the nervous system. It contains a catalytic RING finger domain and NHL domains that facilitate substrate recognition. TRIM2 plays a vital neuroprotective role by mediating the ubiquitination of the neurofilament light subunit (NF-L) and the pro-apoptotic protein BIM (BCL2L11). Clinically, TRIM2 deficiency is a well-established cause of progressive neurodegeneration, characterized by the accumulation of NF-L in axons, leading to juvenile-onset tremor and ataxia. Conversely, TRIM2 dysregulation is also implicated in oncogenesis; by promoting the degradation of BIM, it prevents apoptosis and supports tumor survival in breast and ovarian cancers. This dual role positions TRIM2 as a critical regulator of both neuronal integrity and programmed cell death in malignant contexts. |
