| 背景信息 | Doublecortin like kinase 3 (DCLK3) is a neuron-specific serine/threonine-protein kinase predominantly expressed in the adult forebrain, including the striatum and dentate gyrus. Unlike its paralogs DCLK1 and DCLK2, it lacks a doublecortin domain but facilitates protein localization to the nucleus and regulates peptidyl-serine phosphorylation. DCLK3 modulates transcriptional programs and chromatin remodeling through interactions with the SAGA complex component TADA3 and various zinc finger proteins. Clinically, DCLK3 is recognized for its neuroprotective role in Huntington’s disease. Its expression is significantly reduced in disease models, and this loss heightens striatal vulnerability to mutant huntingtin toxicity. Conversely, the overexpression of DCLK3 has been shown to ameliorate motor deficits and reduce neurodegeneration, establishing it as a critical factor in maintaining neuronal health within the basal ganglia. |
