| 背景信息 | Cilia and flagella associated protein 74 (CFAP74) is a large protein localized to the sperm flagellum and motile cilia, where it forms part of the C1d projection in the central apparatus of the axoneme. It is essential for axoneme assembly, coordinated ciliary beating, and the maintenance of sperm architecture. CFAP74 ensures the integrity of the central pair complex and the proper localization of outer dynein arms, which are required for mucociliary clearance and sperm propulsion. Biallelic loss-of-function mutations in CFAP74 are a well-established cause of Primary Ciliary Dyskinesia 49. This condition manifests as chronic airway disease and male infertility, characterized by subtle ciliary motility defects and impaired fluid flow. |
