| 背景信息 | Ankyrin repeat domain 2 (ANKRD2), also known as ARPP, is a member of the muscle ankyrin repeat protein (MARP) family primarily expressed in cardiac and slow skeletal muscle. It serves as a critical mechanosensor by binding to titin at the sarcomere I-band and translocating to the nucleus in response to mechanical stretch or oxidative stress. In the nucleus, ANKRD2 interacts with transcription factors and regulatory proteins such as p53, PML, and Nkx2.5 to modulate myogenic differentiation and inflammatory signaling. Clinically, ANKRD2 expression is significantly altered in various muscle pathologies; it is upregulated in dilated cardiomyopathy and shows distinct patterns in amyotrophic lateral sclerosis (ALS). Furthermore, defects in its binding to lamin A are associated with Emery-Dreifuss muscular dystrophy, which impairs nuclear import and the cellular response to oxidative stress. |
