| 背景信息 | NSF attachment protein beta (NAPB) is a brain-enriched protein belonging to the soluble N-ethylmaleimide-sensitive fusion attachment protein (SNAP) family. It serves as an essential cofactor for NSF ATPase within the 20S NSF-SNAP-SNARE complex, facilitating the disassembly of SNARE complexes to recycle components for synaptic vesicle fusion and vesicular transport. NAPB also plays a role in AMPA receptor processing through its interaction with GluR2. Clinical relevance is well-established, as loss-of-function mutations in the NAPB gene cause developmental and epileptic encephalopathy 107. This severe neurodevelopmental disorder is characterized by early-onset epilepsy, autism, and significant cognitive impairment, highlighting the protein's critical role in maintaining normal synaptic function. |
