| 背景信息 | SH2 domain containing 4A (SH2D4A) is a cytosolic adaptor protein characterized by a central Src homology 2 (SH2) domain that selectively binds phosphotyrosine motifs. It functions as a negative regulator of signaling by competing with phospholipase C gamma (PLCG) for binding to estrogen receptor alpha (ESR1), thereby inhibiting downstream estrogen-responsive pathways. SH2D4A also facilitates the recruitment of phosphatases, such as protein phosphatase 1, to modulate phosphotyrosine networks and cytoskeletal dynamics. It plays a role in T-cell development and the regulation of cellular proliferation. Clinically, the downregulation of SH2D4A is implicated in the progression of estrogen-driven breast cancer and other hormone-dependent malignancies, where it acts as a putative tumor suppressor. |
