| 背景信息 | Tripartite motif containing 45 (TRIM45) is an E3 ubiquitin-protein ligase characterized by an N-terminal RBCC/TRIM domain, which includes a RING finger, two B-boxes, and a coiled-coil region. This structural arrangement facilitates its ubiquitin ligase activity, specifically enabling the K48-linked polyubiquitination of targets such as TAB2. Through this mechanism, TRIM45 promotes the proteasomal degradation of its substrates, thereby acting as a negative regulator of TLR-mediated inflammatory responses and proteasome-mediated protein catabolism. Additionally, TRIM45 functions as a transcriptional repressor that inhibits Elk-1 and AP-1 within the MAPK signaling pathway. The protein is broadly expressed in both adult and embryonic tissues, localizing to the cytosol, nucleoplasm, and intercellular bridges.
