| 背景信息 | Interleukin 21 (IL21) is a type I four-helix-bundle cytokine belonging to the common γ-chain family, primarily produced by activated CD4+ T cells, including T follicular helper and Th17 cells. It signals through a heterodimeric receptor complex consisting of IL-21R and the common γ chain, activating the JAK1/JAK3–STAT3 pathway to regulate gene expression in various immune cells. IL21 promotes B-cell proliferation, plasma cell differentiation, and immunoglobulin class switching, while also enhancing the cytotoxic activity of CD8+ T cells and NK cells. Additionally, it supports Th17 cell differentiation and modulates dendritic cell activation. Clinically, dysregulated IL21 signaling is implicated in the pathogenesis of autoimmune and inflammatory diseases, such as systemic lupus erythematosus and psoriasis. Due to its ability to bolster antitumor immunity by stimulating cytotoxic lymphocytes, IL21 is also under investigation as a potential cancer immunotherapeutic agent. |
