| 背景信息 | EMAP like 4 (EML4) is a cytoplasmic protein essential for the regulation of microtubule formation, stability, and mitotic spindle organization. It contains several structural domains, including a trimerization domain and a tandem atypical propeller (TAPE) domain, which facilitates tubulin binding and cytoskeletal organization. EML4 is most notably recognized for its pathological role when fused with anaplastic lymphoma kinase (ALK). The resulting EML4-ALK fusion protein undergoes constitutive activation via the EML4 trimerization domain, driving oncogenic signaling through the PI3K/AKT, JAK/STAT, and MEK/ERK pathways. Clinically, the EML4-ALK fusion is a well-established driver in a significant subset of lung adenocarcinomas, as well as some breast and colorectal cancers, making it a primary target for tyrosine kinase inhibitor therapies. |
