| 背景信息 | Leucine zipper transcription factor like 1 (LZTFL1) is a ubiquitously expressed cytoplasmic protein that regulates ciliary protein trafficking and hedgehog signaling. It functions by interacting with the Bardet-Biedl syndrome (BBS) complex, specifically binding to BBS9 to control the ciliary localization of smoothened (SMO). Beyond its role in ciliogenesis, LZTFL1 acts as a tumor suppressor by inhibiting epithelial-to-mesenchymal transition and modulates immune responses by enhancing T cell activation. Clinically, mutations in LZTFL1 are well-established causes of Bardet-Biedl syndrome, characterized by retinal degeneration and obesity. Furthermore, a specific genetic variant of LZTFL1 on chromosome 3 has been identified as a significant risk factor for susceptibility to SARS-CoV-2 infection and the development of severe COVID-19 respiratory failure, highlighting its critical role in both developmental biology and acute viral pathology. |
