| 背景信息 | Tripartite motif containing 36 (TRIM36) is a cytosolic, microtubule-associated RING-type E3 ubiquitin ligase belonging to the TRIM/RBCC family. It is characterized by a RING domain, B-boxes, and a coiled-coil region, which enable it to mediate the ubiquitination and proteasomal degradation of various substrates. TRIM36 plays a critical role in cell-cycle regulation, specifically influencing chromosome segregation, spindle organization, and mitotic cytokinesis by modulating plus-end microtubule dynamics. It is highly expressed in the testis, prostate, and brain, and is essential for early developmental processes, including neural tube closure. Clinically, biallelic loss-of-function variants in TRIM36 are a known cause of autosomal-recessive anencephaly, a severe neural tube defect. Additionally, altered expression of TRIM36 has been implicated in prostate cancer, where it may modulate MAPK/ERK signaling and cellular proliferation. |
