| 背景信息 | Regulator of G protein signaling 18 (RGS18) is a member of the RGS family that acts as a GTPase-activating protein (GAP) for Gαi and Gαq subunits. By accelerating the hydrolysis of GTP to GDP, it effectively terminates G-protein-coupled receptor signaling. RGS18 is highly expressed in megakaryocytes and platelets, where it serves as a critical modulator of platelet reactivity. Its activity is regulated by phosphorylation and subsequent binding to 14-3-3γ, a process that is reversed by cAMP and cGMP signaling pathways to release the protein for G-protein inhibition. Clinically, RGS18 is significant in cardiovascular health; reduced expression of the protein due to specific genetic variants is linked to an increased risk of atherosclerosis and thrombotic events. Knockout models further demonstrate that the absence of RGS18 leads to exaggerated platelet reactivity and heightened susceptibility to thrombosis. |
