| 背景信息 | RAD18 E3 ubiquitin protein ligase (Rad18) is a highly conserved enzyme that plays a fundamental role in postreplication DNA repair and genome stability. It functions by partnering with E2 ubiquitin-conjugating enzymes, such as Rad6, to specifically monoubiquitinate proliferating cell nuclear antigen (PCNA) at lysine 164. This modification serves as a critical signal to recruit translesion DNA synthesis (TLS) polymerases to sites of UV-induced damage, preventing replication fork collapse. Rad18 also acts as a scaffolding protein, recruiting the SMC5/6 complex to double-strand breaks and promoting homologous recombination. Furthermore, it facilitates the activation of the Fanconi anemia pathway by ubiquitylating FANCD2 and FANCI. Through these mechanisms, Rad18 ensures the bypass of DNA lesions and maintains chromosomal integrity during the G2/M checkpoint. |
