背景信息Diaphanous related formin 3 (DIAPH3) is a member of the formin homology protein family, primarily functioning as an actin nucleation and elongation factor essential for the assembly of F-actin structures such as actin cables and stress fibers. It plays a critical role in cellular processes including cytokinesis, cell shape changes, adhesion, and division by promoting actin polymerization in both the cytoplasm and nucleus, often in a Rho-dependent manner. DIAPH3 also interacts with microtubules to stabilize their structure and is involved in the regulation of selenoamino acid metabolism through interactions with ribosomal proteins, potentially influencing selenoprotein expression. Disease associations include its implication in auditory neuropathy, autosomal dominant 1 (AUNA1), and its overexpression has been linked to the progression of certain cancers, such as pancreatic cancer, by altering cytoskeletal dynamics and cellular metabolism.
