| 背景信息 | Ring finger protein 19A, RBR E3 ubiquitin protein ligase (RNF19A) is an E3 ligase belonging to the RBR family that regulates protein degradation pathways associated with neuroprotection and tumor growth. It facilitates the transfer of ubiquitin from E2 enzymes, such as UBE2L3 and UBE2L6, to specific substrates including synphilin-1 and pathogenic variants of SOD1, promoting their proteasomal clearance. RNF19A is localized to Lewy bodies, the characteristic protein inclusions found in neurodegenerative disorders. Clinically, RNF19A is implicated in the pathogenesis of Parkinson's disease, amyotrophic lateral sclerosis (ALS), and Gordon Holmes syndrome. Additionally, its overexpression in non-small cell lung cancer has been linked to the degradation of p53, thereby promoting uncontrolled tumor cell proliferation. |
