| 背景信息 | WD repeat domain 70 (WDR70) is a nuclear scaffolding protein characterized by multiple WD40 repeats that form a beta-propeller structure. It functions as a critical subunit of the CRL4 ubiquitin ligase complex, where it coordinates the DNA damage response and maintains genomic stability. WDR70 localizes to sites of DNA double-strand breaks and regulates the transcription of essential repair genes, including BRCA1, BRCA2, and RAD51, thereby promoting both homologous recombination and non-homologous end joining. Furthermore, it interacts with the RNF20/40 E3 ligase to enhance histone H2BK120 monoubiquitylation, which facilitates downstream H3K79 methylation and the expression of POLE3. Through these mechanisms, WDR70 acts as a central regulator of chromatin modifications and DNA repair pathways. |
