| 背景信息 | Interleukin 36 beta (IL-36β), also known as IL-1 family member 8 (IL1F8), is a cytokine belonging to the interleukin-1 superfamily. It is encoded by the IL36B gene located on chromosome 2, forming part of a cytokine gene cluster. Structurally, IL-36β contains a conserved 12-stranded β-trefoil fold similar to IL-1α and IL-1β, and exists in two splice variants encoding distinct isoforms. IL-36β acts as an agonist for the IL-36 receptor (IL-36R), activating downstream signaling pathways such as NF-κB and MAPK. These pathways promote pro-inflammatory responses, including the secretion of IL-6 and IL-8, and are involved in immune cell activation and antigen presentation. Predominantly expressed in epithelial tissues like skin and lungs, IL-36β plays a key role in regulating inflammation. Dysregulation of IL-36β has been implicated in inflammatory diseases such as psoriasis, rheumatoid arthritis, and inflammatory bowel disease. Its overactivation contributes to chronic inflammation and autoimmune pathologies. |
