| 背景信息 | Intersectin 2 (ITSN2) is a large cytoplasmic scaffold and adaptor protein that regulates clathrin-mediated endocytosis and actin cytoskeletal dynamics. It contains multiple functional domains, including EH, coiled-coil, and SH3 domains, which enable extensive protein-protein interactions. ITSN2 facilitates the formation and maturation of clathrin-coated vesicles and is particularly involved in T cell antigen receptor (TCR) endocytosis. By interacting with partners such as Cdc42 and WASp, it links endocytic membranes to the actin assembly machinery. Through these mechanisms, ITSN2 integrates endocytosis with intracellular signaling pathways and vesicle trafficking. Dysregulation of intersectin proteins has been associated with tumor progression in various cancers and with neurodegenerative conditions such as Down syndrome and Alzheimer’s disease. |
