| 背景信息 | EH domain containing 3 (EHD3) is a membrane-associated protein of the EHD family that regulates endocytic recycling and membrane tubulation. It facilitates the transport of cargo from early endosomes to the endocytic recycling compartment and the plasma membrane. Structurally, EHD3 contains an N-terminal dynamin-like G domain, a central coiled-coil for oligomerization, and a C-terminal EH domain that binds calcium. In cardiac myocytes, EHD3 is essential for the proper targeting of ion transporters; it directly interacts with ankyrin-B to control the surface expression of the Na+/Ca2+ exchanger (NCX1). Clinically, EHD3 deficiency is linked to cardiac conduction disorders and arrhythmogenesis. The loss of EHD3 leads to impaired NCX1 membrane localization and altered electrical excitability, predisposing the heart to arrhythmias. |
