| 背景信息 | CD274 encodes the protein PD-L1, which is an immune inhibitory receptor ligand expressed by various cells, including T cells, B cells, and tumor cells. The interaction of PD-L1 with its receptor PD-1 on activated T cells transmits an inhibitory signal that reduces T cell proliferation and activation, helping to maintain immune homeostasis. However, in the tumor microenvironment, this interaction allows tumor cells to evade immune surveillance and escape destruction by cytotoxic T cells. Disorders in the expression and regulation of CD274/PD-L1 have been associated with various diseases. Overexpression of PD-L1 in tumors is linked to immune escape and worse prognosis in many cancers. Conversely, downregulation of PD-L1 has been observed in autoimmune diseases like systemic lupus erythematosus, potentially contributing to loss of peripheral tolerance. Overall, CD274/PD-L1 plays a critical role in modulating immune responses, with implications for both cancer and autoimmunity. |
