| 背景信息 | MacroH2A.2 histone (MACROH2A2) is a replication-independent histone variant that replaces conventional H2A in specific nucleosomes to facilitate transcriptional repression and stabilize chromosomal architecture. It features a unique tripartite structure consisting of a histone fold domain, an intrinsically disordered linker that enhances chromatin condensation, and a C-terminal macrodomain. Unlike other macroH2A variants, the macrodomain of MACROH2A2 is enzymatically inert and does not bind ADP-ribose or respond to PARP1 signaling, suggesting its primary role is structural rather than metabolic. MACROH2A2 is particularly involved in the maintenance of X chromosome inactivation and the regulation of nuclear organization. Its presence in the chromatin fiber limits plasticity and reduces DNA damage-induced relaxation, thereby contributing to the long-term silencing of developmentally regulated genes. |
