| 背景信息 | Pim-2 proto-oncogene, serine/threonine kinase (PIM2) is a constitutively active cytoplasmic kinase that serves as a critical regulator of cell survival and proliferation. It functions by phosphorylating key substrates, including BAD, MYC, and 4E-BP1, thereby inhibiting apoptosis and enhancing cap-dependent translation through an mTORC1-independent pathway. PIM2 also supports NF-κB-dependent survival signaling and maintains mitochondrial potential, providing resistance to cytotoxic stimuli and growth factor withdrawal. Clinically, PIM2 is frequently overexpressed in various hematologic malignancies and solid tumors, where it acts as an oncogenic driver by promoting metabolic reprogramming, enhanced glycolysis, and sustained proliferative signaling. |
