| 背景信息 | Guanosine monophosphate reductase 2 (GMPR2) is a human enzyme that catalyzes the irreversible, NADPH-dependent reductive deamination of guanosine monophosphate (GMP) to inosine monophosphate (IMP). This reaction is critical for the salvage of purine nucleotides and the maintenance of intracellular balance between adenine and guanine nucleotides. Structurally, the enzyme forms a homotetramer characterized by an eight-stranded barrel core stabilized by hydrophobic residues and specific disulfide bonds. GMPR2 plays a significant role in cellular differentiation, notably promoting monocytic differentiation in hematopoietic lineages. Beyond its metabolic functions, the enzyme is implicated in the suppression of melanoma through GTP depletion and has been associated with the progression of certain malignancies, such as urethra cancer. |
