| 背景信息 | Solute carrier family 17 member 6 (SLC17A6), also known as vesicular glutamate transporter 2 (VGLUT2), is a membrane protein that mediates electrogenic uptake of L‑glutamate and phosphate into synaptic vesicles of excitatory neurons, driven by the proton electrochemical gradient generated by vesicular H+-ATPase. It also transports chloride, potassium, sodium and phosphate, functioning as a glutamate uniporter, chloride channel, and K+/H+ antiporter, thereby shaping vesicle acidification and sustaining vesicular glutamate loading. SLC17A6 is highly expressed in brain, localizing predominantly to synaptic vesicle membranes, and participates in neuronal signaling pathways involving transport of inorganic ions and amino acids. Clinically, SLC17A6 has been associated with rabies and arthrogryposis, distal, type 2A, and is investigated in broader neuropsychiatric and neurodevelopmental contexts given its central role in excitatory neurotransmission and hippocampal and brain development. An important paralog is SLC17A7 (VGLUT1). |
