| 背景信息 | (PDGFRA) is a type III receptor tyrosine kinase that serves as a critical regulator of cell proliferation, migration, survival, and differentiation. It features five extracellular immunoglobulin-like domains for binding PDGF isoforms, a transmembrane helix, and a split cytoplasmic kinase domain. Upon ligand binding, the receptor undergoes dimerization and autophosphorylation on multiple tyrosine residues, relieving autoinhibition and recruiting SH2-domain-containing proteins such as PI3K, PLCγ, and Src. These interactions initiate downstream signaling through the MAPK/ERK and PI3K/AKT pathways. Primarily expressed in mesenchymal cells, fibroblasts, and glia, it supports embryonic development and vascular integrity. Clinically, gain-of-function mutations or fusions, such as FIP1L1-PDGFRA, are well-established drivers of gastrointestinal stromal tumors, chronic eosinophilic leukemia, and inflammatory fibrosis. |
