| 背景信息 | Fetuin B (Fetuin-B) is a secreted serum glycoprotein belonging to the cystatin superfamily of cysteine protease inhibitors. It serves as a highly potent and selective inhibitor of astacin-family metallopeptidases, with its primary physiological target being ovastacin. This interaction is critical for female fertility, as Fetuin-B prevents the premature hardening of the zona pellucida by inhibiting ovastacin-mediated cleavage of ZP2 before fertilization occurs. Structurally, the protein utilizes a unique inhibition mechanism involving tandem cystatin-like domains and a disulfide-linked loop that occludes the catalytic zinc site of its target enzymes. Beyond its role in reproduction, Fetuin-B is involved in broader physiological processes, including the regulation of osteogenesis, insulin receptor signaling, and the systemic inflammatory response. |
