| 背景信息 | CD300a molecule (CD300A) is a transmembrane glycoprotein and inhibitory receptor expressed on various leukocytes. It functions as a negative regulator of immune signaling through four cytoplasmic ITIM domains that recruit protein tyrosine phosphatases, including SHP-1, SHP-2, and SHIP. This recruitment leads to the downregulation of cytolytic activity in natural killer cells, inhibition of mast cell degranulation, and suppression of Toll-like receptor signaling. In the context of hematologic malignancies, CD300A acts as a tumor promoter by modulating the PECAM1/AKT/mTOR signaling pathway, which enhances cell proliferation and inhibits apoptosis. Clinically, high expression of CD300A is a well-established prognostic marker in acute myeloid leukemia, where it is associated with poor clinical outcomes and represents a potential therapeutic target. |
