| 背景信息 | MutL homolog 3 (MLH3) is a highly conserved DNA mismatch repair protein that forms the MutLγ heterodimer with MLH1. This complex plays a dual role in somatic mismatch repair and meiotic recombination. MLH3 possesses a latent endonuclease function activated by Holliday junctions, which is essential for crossover formation during meiosis. Structurally, it features a transducer domain and a ribosomal S5-like domain that facilitate the protein compaction and oligomerization necessary for recombination. MLH3 also participates in class switch recombination and somatic hypermutation in B cells. Clinically, germline mutations in MLH3 are linked to hereditary nonpolyposis colorectal cancer type 7, while somatic mutations are frequently associated with microsatellite instability in various tumors. |
