| 背景信息 | Transcriptional repressor GATA binding 1 (TRPS1) is a zinc finger transcription factor that specifically binds to GATA sequences and represses the expression of GATA-regulated genes at selected sites and stages in vertebrate development. The protein contains nine zinc-finger domains, including a single GATA-type DNA binding domain flanked by two potential regulatory domains. TRPS1 functions downstream of BMP7 and GDF5 signaling pathways, regulating key developmental processes including chondrocyte condensation, proliferation, and maturation. It plays crucial roles in bone development, hair follicle morphogenesis, kidney tubule formation, and phalangeal joint development by targeting genes such as Sox9, Pthrp, Stat3, and Runx2, and by directly interacting with Gli3. TRPS1 is also implicated in tumorigenesis, invasion, and metastasis in prostate and breast cancers. Mutations in the TRPS1 gene are associated with tricho-rhino-phalangeal syndrome (TRPS) types I and III, which are autosomal dominant skeletal disorders characterized by craniofacial and skeletal abnormalities. |
