| 背景信息 | CD84 molecule (CD84) is a SLAM family membrane glycoprotein that functions as a homophilic adhesion molecule on various immune cells, including B cells, T cells, monocytes, and platelets. It mediates ligand-receptor recognition through self-binding of its extracellular immunoglobulin-like domains, facilitating stable interactions within the immunological synapse. Upon homophilic engagement, CD84 recruits adapter proteins such as SH2D1A (SAP) and SH2D1B (EAT-2) to enhance interferon-gamma secretion and T-cell proliferative responses. It plays a vital role in T follicular helper cell function, germinal center formation, and the maintenance of B-cell tolerance. Clinically, CD84 dysfunction is associated with X-linked lymphoproliferative disease, as its cytoplasmic tail interacts directly with the protein product of the disease-associated gene. |
