| 背景信息 | Methionine sulfoxide reductase A (MSRA) is a thioredoxin-dependent antioxidant enzyme that stereospecifically reduces the S-epimer of methionine sulfoxide back to methionine. This catalytic activity is essential for repairing proteins damaged by reactive oxygen species and maintaining cellular redox homeostasis. MSRA exists in multiple isoforms that localize to the cytosol, mitochondria, and nucleus. Its mechanism involves a three-step ping-pong process utilizing essential cysteine residues to perform a nucleophilic attack on the substrate, followed by thioredoxin-mediated regeneration of the active site. By scavenging oxidants and repairing oxidative damage, MSRA serves as a primary defense mechanism against oxidative stress and may also participate in redox-based cell signaling. |
