| 背景信息 | F-box and leucine rich repeat protein 5 (FBXL5) is a substrate recognition component of the SCF ubiquitin ligase complex that serves as a master regulator of cellular iron homeostasis. It mediates the ubiquitination and subsequent proteasomal degradation of iron regulatory protein 2 (IRP2) in an iron- and oxygen-dependent manner. FBXL5 features a hemerythrin-like domain that acts as a sensor, stabilizing the protein only when iron and oxygen levels are sufficient. In addition to IRP2, it targets other substrates such as SNAI1 and NABP2. Clinically, FBXL5 deficiency is linked to systemic iron overload and hematopoietic stem cell exhaustion, contributing to myelodysplastic syndrome-like defects. It also plays a protective role against hepatocarcinogenesis by suppressing oxidative stress and DNA damage in the liver. |
