| 背景信息 | F-box and WD repeat domain containing 11 (FBXW11), also known as beta-TrCP2, is the substrate-recognition subunit of the SCF (SKP1-CUL1-F-box) E3 ubiquitin ligase complex. It utilizes C-terminal WD40 repeats to recognize phosphorylated degrons on various target proteins, including beta-catenin, IkBa, and PER1/2. By mediating the ubiquitin-dependent proteolysis of these substrates, FBXW11 regulates critical signaling pathways such as Wnt, NF-kappaB, and the circadian clock. Clinically, FBXW11 is overexpressed in acute lymphoblastic leukemia (ALL), where it promotes leukemic cell proliferation and tumor formation by activating NF-kappaB and beta-catenin/TCF signaling. This suggests an oncogenic role for FBXW11 in hematologic malignancies and identifies it as a potential therapeutic target. |
