| 背景信息 | Adaptor protein, phosphotyrosine interacting with PH domain and leucine zipper 1 (APPL) is a multifunctional endosomal adaptor protein that coordinates receptor trafficking and intracellular signaling. It localizes to a distinct population of Rab5-positive early endosomes via its N-terminal BAR and PH domains. The protein's C-terminal phosphotyrosine-binding domain facilitates interactions with key signaling molecules, including Akt2, TrkA, and AdipoR1, integrating pathways for cell survival, adhesion, and migration. APPL1 also undergoes endosomal-to-nuclear translocation to influence gene transcription. Furthermore, it regulates integrin trafficking and focal adhesion turnover by associating with Arf6 effectors. These functions make it a central hub for coupling endocytic membrane dynamics with complex cellular signaling cascades. |
