| 背景信息 | Histone deacetylase 9 (HDAC9) is a class IIa enzyme that regulates gene expression by deacetylating lysine residues on core histones, leading to chromatin condensation and transcriptional repression. It features a conserved C-terminal catalytic domain and an N-terminal regulatory domain that facilitates interactions with various transcription factors. HDAC9 shuttles between the nucleus and cytoplasm in a phosphorylation-dependent manner, a process regulated by 14-3-3 protein binding. The enzyme plays critical roles in diverse biological processes, including the inhibition of muscle differentiation through MEF2 interaction and the regulation of bone homeostasis by suppressing osteoclastogenesis. Clinically, HDAC9 is associated with cardiac hypertrophy, atherosclerosis, and neurodevelopmental disorders such as autism. Genetic variants and altered expression levels are also linked to changes in bone mineral density and inflammatory vascular diseases. |
