| 背景信息 | Proteasome activator subunit 2 (PSME2), also known as PA28β, is a crucial component of the immunoproteasome, a specialized form of the proteasome involved in processing viral and tumor antigens for presentation by MHC class I molecules, thus playing a vital role in immune surveillance. Structurally, PSME2 forms part of the 11S regulator complex, consisting of heterohexameric rings of alpha and beta subunits, which bind to the 20S proteasome core to induce conformational changes that modulate substrate entry and proteolytic activity. This modulator function extends to interactions that can suppress chymotrypsin-like activity independently of the 26S proteasome. Dysregulation of PSME2 expression has been linked to various cancers; downregulation correlates with increased invasiveness in gastric adenocarcinoma and esophageal squamous cell carcinoma. In contrast, PSME2 acts as a tumor suppressor in breast cancer and osteosarcoma, highlighting its potential as a prognostic marker and therapeutic target. Additionally, PSME2's circadian regulation suggests roles in cellular proteostasis and oxidative stress response that may influence aging and disease. |
