背景信息Aspartyl aminopeptidase (DNPEP) is a zinc-dependent metalloaminopeptidase belonging to the M18 family, characterized by its strict specificity for acidic N-terminal amino acids, specifically preferring aspartic acid over glutamic acid. The enzyme exhibits a complex dodecameric structure composed of six dimers, forming a tetrahedral arrangement with a central cavity that houses twelve active sites. Each active site contains a binuclear zinc center that coordinates substrates through a hydroxamate-like mechanism, while a restricted P1 pocket enforces the preference for acidic residues. DNPEP is primarily involved in the intracellular processing of peptides, contributing to the final stages of protein degradation and the recycling of amino acids. Its highly specialized catalytic mechanism and structural organization allow for precise control over the cleavage of peptide bonds at the N-terminus of its substrates.
