| 背景信息 | Leucine rich repeat and fibronectin type III domain containing 2 (LRFN2), also known as SALM1, is a type I transmembrane glycoprotein essential for neuronal development and synaptic function. Its extracellular region contains leucine-rich repeat, immunoglobulin-like, and fibronectin type III domains, while its intracellular C-terminus features a PDZ-binding domain. LRFN2 localizes to the plasma membrane and postsynaptic density, where it promotes neurite outgrowth and enhances the cell surface expression of NMDA receptor subunits GRIN1 and GRIN2A. In the retina, it is crucial for the formation of invaginating ribbon synapses at cone pedicles, facilitating normal synaptic signaling. Its role in modulating chemical synaptic transmission and postsynapse organization makes it a key regulator of central nervous system connectivity and the structural integrity of neuronal circuits. |
