| 背景信息 | ALK receptor tyrosine kinase (ALK) is a critical regulator of nervous system development and neuronal differentiation. This receptor features an extracellular domain with unique MAM and LDLa motifs, which bind the ligands ALKAL1 and ALKAL2 to induce receptor dimerization and autophosphorylation. Activation of ALK triggers several downstream signaling cascades, including the MAPK-ERK, PI3K-AKT, and JAK-STAT pathways, which govern retinal axon targeting, synapse development, and cognitive functions. Clinically, ALK is a well-known oncogene; gain-of-function mutations and chromosomal translocations are primary drivers of pediatric neuroblastoma and various other malignancies. Notably, the EML4-ALK fusion protein leads to constitutive, ligand-independent kinase activity, promoting uncontrolled cell proliferation and survival in non-small cell lung cancer and other solid tumors. |
