| 背景信息 | RuvB like AAA ATPase 1 (RuvB Like 1), also known as Pontin or TIP49, is an evolutionarily conserved member of the AAA+ ATPase family. It exhibits single-stranded DNA-stimulated ATPase and DNA helicase activities, typically functioning as part of heterohexameric or dodecameric rings with its paralog RUVBL2. The protein is an integral component of several multisubunit complexes, including NuA4, INO80, and SWR1, where it facilitates chromatin remodeling, transcriptional regulation, and DNA repair. RuvB Like 1 interacts with key oncogenic factors such as c-Myc and β-catenin to support cell proliferation. Clinically, mutations in RUVBL1 are associated with Common Variable Immunodeficiency 15 and neurodevelopmental disorders characterized by progressive spasticity and brain abnormalities, reflecting its fundamental role in genomic stability and gene expression. |
