| 背景信息 | Nuclear distribution C, dynein complex regulator (NUDC) is a conserved protein essential for cellular processes including mitosis, neurogenesis, and nuclear migration. Structurally, NUDC contains a core CS domain resembling small heat shock proteins and co-chaperones, with an N-terminal coiled-coil motif mediating homodimerization. It functions as a chaperone stabilizing target proteins and enhancing their folding, especially in relation to dynein complex regulation. NUDC interacts with the dynein intermediate chain, preventing its aggregation and promoting dynein stability critical for proper spindle formation and chromosome segregation during mitosis. Its expression and phosphorylation increase during mitosis, highlighting its role in cell division. Deficiency in NUDC causes mitotic arrest, defective spindle function, and cell death. In disease contexts, NUDC dysfunction has been implicated in neurodevelopmental disorders and cancer, due to its key role in mitotic progression and maintenance of cellular architecture. |
