| 背景信息 | V-set and immunoglobulin domain containing 4 (VSIG4), also known as CRIg, is a type I transmembrane protein and immune checkpoint exclusively expressed on tissue-resident macrophages. It functions as a phagocytic receptor that binds complement fragments C3b and iC3b to facilitate pathogen clearance and inhibit the alternative complement pathway. VSIG4 also plays a significant role in maintaining immune tolerance by potently inhibiting T-cell proliferation and IL-2 production. Clinically, while VSIG4 helps curb autoimmune and inflammatory conditions, its overexpression in high-grade gliomas is a well-established driver of immune escape, promoting tumor invasion and T-cell dysfunction. In breast cancer, its expression levels are modulated by microRNAs and serve as a prognostic indicator, highlighting its complex role in both host defense and tumor progression. |
