背景信息Phosphoinositide kinase, FYVE-type zinc finger containing (PIKFYVE) is an enzyme encoded by the PIKFYVE gene in humans, which plays a crucial role in phosphoinositide metabolism. It primarily phosphorylates phosphatidylinositol 3-phosphate (PtdIns3P) to produce phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2) and phosphatidylinositol 5-phosphate (PtdIns5P). PIKFYVE is a large protein with multiple functional domains and is expressed in several spliced forms. Its enzymatic activity is essential for vesicular dynamics, affecting trafficking pathways through the endosomal system. Impaired PIKFYVE activity leads to lysosome enlargement and cytoplasmic vacuolation due to disrupted PtdIns(3,5)P2 synthesis and lysosome fission. Mutations in PIKFYVE are associated with Francois-Neetens corneal fleck dystrophy and have been implicated in pathogen invasion, including HIV and Salmonella replication, as well as type 2 diabetes through the inhibition of insulin-regulated glucose uptake. PIKFYVE also interacts with other proteins to form a complex that tightly controls PtdIns(3,5)P2 levels, highlighting its importance in cellular processes.
