| 背景信息 | DIS3 homolog, exosome endoribonuclease and 3'-5' exoribonuclease (DIS3) is a conserved hydrolytic enzyme that serves as a catalytic subunit of the RNA exosome complex, responsible for 3' to 5' degradation and processing of single-stranded RNA. It exhibits both processive exoribonuclease and endoribonuclease activities, with exonuclease function mediated by four conserved aspartic acid residues coordinating magnesium ions in its catalytic center, while the PIN domain provides endonucleolytic cleavage. DIS3 can unwind structured RNA substrates and its activity is modulated allosterically when part of the exosome complex. The protein is essential for RNA homeostasis and degradation of RNA intermediates in the nucleus and cytoplasm. Mutations in DIS3 are implicated in multiple myeloma, correlating with poorer patient survival, and often co-occur with chromosomal deletion del(13q). These mutations affect RNA processing and contribute to disease progression, underscoring DIS3's critical role in RNA metabolism and its impact on cancer pathogenesis. |
