| 背景信息 | (NRXN3) is a presynaptic cell adhesion molecule belonging to the neurexin family that plays a critical role in organizing nanoscale synaptic architecture. It exists as long α-isoforms and shorter β-isoforms, which undergo extensive alternative splicing to modulate ligand binding. NRXN3 forms trans-synaptic bridges with postsynaptic partners, including neuroligins, LRRTMs, cerebellins, and GluDs, to stabilize AMPA and NMDA receptor nanocolumns. Mechanistically, it regulates presynaptic release probability through its intracellular C-terminal PDZ-binding motifs, which recruit synaptotagmin and RIM. By controlling excitatory and inhibitory synapse strength and plasticity, NRXN3 ensures precise trans-synaptic alignment and connectivity. Genetic variants or dysregulation of NRXN3 are associated with neurodevelopmental and psychiatric conditions, including autism spectrum disorders, schizophrenia, and substance dependence involving nicotine and alcohol. |
