| 背景信息 | Mitogen-activated protein kinase kinase kinase kinase 5 (MAP4K5) is a serine/threonine protein kinase belonging to the SPS1/STE20 family. It functions as a key component of the MAP kinase signaling cascade, relaying signals from G proteins to cytosolic MAP kinases to regulate intracellular transduction and protein phosphorylation. MAP4K5 acts upstream of the JUN N-terminal (JNK) pathway and is involved in cellular responses to environmental stress, as well as the TGF-beta and angiopoietin-like protein 8 regulatory pathways. It interacts with adapter proteins such as CRKL and TRAF2 to coordinate these signaling events. Clinically, MAP4K5 is associated with congenital myasthenic syndrome 3B and acute myeloid leukemia featuring NPM1 mutations. |
