| 背景信息 | Ankyrin repeat and SOCS box containing 3 (ASB3) is a substrate-recognition component of an SCF-like E3 ubiquitin ligase complex. It features multiple ankyrin repeats and a SOCS box domain, which facilitates the coupling of target proteins to the elongin B/C complex for proteasomal degradation. ASB3 plays a significant role in modulating immune and inflammatory signaling by ubiquitinating key targets, including TNFRSF1B (TNF receptor II), TRAF6, and MAVS. Through the K48-linked polyubiquitination of TRAF6, ASB3 promotes NF-κB activation and the production of proinflammatory cytokines. Clinically, ASB3 is upregulated in inflammatory bowel disease (IBD), where it contributes to the aggravation of colitis by disrupting intestinal barrier integrity and dysregulating the gut microbiota. Its role in downregulating antiviral immunity highlights its importance in immune homeostasis. |
