| 背景信息 | Tripartite motif containing 17 (TRIM17) is an E3 ubiquitin ligase that plays a pivotal role in neuronal apoptosis, selective autophagy, and the regulation of cell proliferation. It contains characteristic RING, B-box, and coiled-coil domains, localizing primarily to cytoplasmic bodies. TRIM17 initiates neuronal death by mediating the ubiquitination and subsequent degradation of the anti-apoptotic protein MCL1. Furthermore, it inhibits autophagic degradation by modulating the assembly of the MCL1-BECN1 complex and negatively regulates cell proliferation through the degradation of ZWINT. Beyond these roles, TRIM17 regulates NFAT transcription factors and alpha-synuclein transcription in neurons. Clinically, TRIM17 is associated with Autism Spectrum Disorder and Opitz GBBB Syndrome, reflecting its importance in neurological development and cellular homeostasis. |
