| 背景信息 | CDC42 binding protein kinase beta (CDC42BPB), also known as MRCK beta, is a serine/threonine-protein kinase that serves as a key downstream effector of the CDC42 small GTPase. It regulates actin cytoskeletal reorganization, cell migration, and invasion by phosphorylating substrates such as MYPT1 and MLC2, thereby modulating actomyosin contractility. Additionally, CDC42BPB facilitates the activation of LIMK1 to regulate cofilin phosphorylation in lamellipodia, which is essential for retrograde actomyosin flow and cellular motility. It is expressed in various tissues, including the brain, muscle, and gonads. Clinically, biallelic loss-of-function variants in CDC42BPB are associated with Chilton-Okur-Chung neurodevelopmental syndrome (CHOCNS), a condition characterized by intellectual disability, seizures, and hypotonia. |
